D1.2 - Application specific cartridge prototype, protocols and isolated ENPs for Two-Step ENP isolation Methods
The SMART-NANO consortium developed several protocols for the pre-treatment of ENP-containing samples prior to analysis. Several methods for matrix simplification have been tested and bench-top experimental protocols were developed and optimized. Working sample preparation protocols have been achieved for challenging samples, such as TiO2 NP in sunscreen lotions, SiO2 NP in milk, and Ag NP in environmental samples. For the in biota samples a protocol was developed which allows the AF4 separation of the supernatant cell lysed component of bacterial and diatoms cell cultures. These sample preparation methods thus far have proved absolutely essential to the successful measurement of particle size distribution in complex samples. Based on the expertise gained on the bench, a sample preparation cartridge was produced and tested. The results show that the cartridge can be used both as for the matrix simplification in the case of complex samples (such as sunscreen lotions) and as for the up-concentration of diluted environmental samples.
